ABSTRACT

Mushrooms are a group of fungi with great diversity and ultra-accelerated metabolism. As a consequence, mushrooms have developed a protective mechanism consisting of high concentrations of antioxidants such as selenium, polyphenols, β-glucans, ergothioneine, various vitamins and other bioactive metabolites. The mushrooms of the Pleurotus genus have generated scientific interest due to their therapeutic properties, especially related to risk factors connected to the severity of coronavirus disease 2019 (COVID-19). In this report, we highlight the therapeutic properties of Pleurotus mushrooms that may be associated with a reduction in the severity of COVID-19: antihypertensive, antihyperlipidemic, antiatherogenic, anticholesterolemic, antioxidant, anti-inflammatory and antihyperglycemic properties. These properties may interact significantly with risk factors for COVID-19 severity, and the therapeutic potential of these mushrooms for the treatment or prevention of this disease is evident. Besides this, studies show that regular consumption of Pleurotus species mushrooms or components isolated from their tissues is beneficial for immune health. Pleurotus species mushrooms may have a role in the prevention or treatment of infectious diseases either as food supplements or as sources for pharmacological agents.

ABSTRACT

Abstract The aim of this research was to evaluate the efficiency of aqueous alkali-treated Brachiaria straw for the cultivation of appropriate species of oyster mushroom. The substrate used in the cultivation of various Pleurotus spp. was soaked for 20 min by using two different procedures: (i) 0.5-2.0% Ca(OH)2 in 100 L water, and (ii) 50-250 L water. As a result, 1% Ca(OH)2 dissolved in 100 L water and 3.5 kg of Brachiaria straw presented the best production. The most suitable species for the application of the present method were P. pulmonarius and P. sapidus. The success of this technique is directly related to the concentration of Ca(OH)2 and water, the species, and the origin and quality of raw material used as the substrate in the production of oyster mushroom.

ABSTRACT

The consumption of fungal- and plant-derived non-starch polysaccharides (NSP) have been associated with reduced risk of cardiovascular diseases and cancer. In addition to promote physiochemical effects on the gastrointestinal tract and serve as substrate for the intestinal microbiota to produce short-chain fatty acids, NSP can interact with immune system cells including macrophages, which are crucial for tissue repair, lipid metabolism and host defense against foreign substances and pathogens. However, the effects of NSP in macrophages depends on their structure. Recently, it was showed that the chayote (Sechium edule) and the fungus Pleurotus albidus are promising sources of NSP with potential immunomodulatory effects in macrophages. In this study, it was explored the effects of cooking on the composition of NSP from chayote and evaluated their biological effects in macrophages. Furthermore, it was optimized a method for the extraction of mushroom NSP and characterized the structure and biological effects of NSP from P. albidus in macrophages. Results showed that the NSP from chayote pulp regulate cytokine secretion and phagocytosis by macrophages, and minor changes in composition during cooking influences their effects in macrophages. Furthermore, NSP from chayote induces cholesterol efflux and inhibits the expression of genes required for NLRP3 inflammasome activation in macrophages previously exposed to cholesterol crystals. Then, it was showed that the optimized method for the extraction of NSP from mushroom reduces by up to half the extraction time commonly required. Furthermore, results showed that P. albidus is source of easily extractable glucans with biological effects in macrophages. Results also suggest that glucans from P. albidus inhibit lipid-induced inflammation and foam-cell formation at distinct levels, with significant effects on NLRP3 inflammasome activation. Taken together, the results suggest that the benefits of chayote NSP is beyond their physical properties on the gastrointestinal tract, and that the P. albidus NSP offers potential health benefits that might be of relevance as a functional food ingredient


O consumo de polissacarídeos não-amido (PNA) de fungos e plantas tem sido associado a redução do risco de doenças cardiovasculares. Além de promoverem efeitos físicos no trato gastrointestinal e serem utilizados como substratos pela microbiota intestinal, os PNA podem interagir com células do sistema imune, como macrófagos, cruciais no reparo tecidual, metabolismo lipídico, e na defesa do organismo contra patógenos. Entretanto, os efeitos em macrófagos dependem da estrutura do PNA. Recentemente, foi observado que o chuchu (Sechium edule) e o fungo Pleurotus albidus são fontes de PNA com potencial efeito sobre macrófagos. Assim, foram avaliados os efeitos dos PNA do chuchu fresco e cozido em macrófagos. Além disso, foi otimizado um método para extração de polissacarídeos de cogumelo, e avaliada a estrutura e os efeitos biológicos dos PNA do P. albidus em macrófagos. Foi observado que os PNA do chuchu regulam a secreção de citocinas e o processo de fagocitose por macrófagos, e alterações na composição de PNA durante o cozimento tem um impacto em seus efeitos biológicos. Além disso, os PNA do chuchu induzem o efluxo de colesterol e regulam a expressão de genes necessários para a ativação do inflamassoma NLRP3 em macrófagos previamente tratados com cristais de colesterol. Também foi demonstrado que o método otimizado de extração de PNA de cogumelos reduz em até pela metade o tempo de extração normalmente empregado. Além disso, foi verificado que o P. albidus é fonte para extração de glicanos com efeitos em macrófagos. Os resultados também sugerem que os glicanos obtidos do P. albidus inibem em diferentes níveis a inflamação induzida por lipídeos e a formação de células espumosas, com efeitos significativos sobre a ativação do inflamassoma NLRP3. Tais diferenças parecem estar associadas à estrutura dos glicanos. Por fim, os resultados sugerem que os benefícios dos PNA do chuchu estão além dos seus efeitos físicos sobre o trato gastrointestinal, e que os PNA do P. albidus promovem benefícios que podem ser relevantes para explorar sua utilização como um alimento ou fonte para extração de ingredientes funcionais

Pleurotus ostreatus is able to bioaccumulate several metals in its cell structures; however, there are no reports on its capacity to bioaccumulate iron. The objective of this study was to evaluate cultivation variables to increase iron bioaccumulation in P. ostreatus mycelium. A full factorial design and a central composite design were utilized to evaluate the effect of the following variables: nitrogen and carbon sources, pH and iron concentration in the solid culture medium to produce iron bioaccumulated in mycelial biomass. The maximum production of P. ostreatus mycelial biomass was obtained with yeast extract at 2.96 g of nitrogen L−1 and glucose at 28.45 g L−1. The most important variable to bioaccumulation was the iron concentration in the cultivation medium. Iron concentration at 175 mg L−1 or higher in the culture medium strongly inhibits the mycelial growth. The highest iron concentration in the mycelium was 3500 mg kg−1 produced with iron addition of 300 mg L−1. The highest iron bioaccumulation in the mycelium was obtained in culture medium with 150 mg L−1 of iron. Iron bioaccumulation in P. ostreatus mycelium is a potential alternative to produce non-animal food sources of iron.

ABSTRACT

Polysaccharides with medicinal properties can be obtained from fruiting bodies, mycelium and culture broth of several fungus species. This work was carried out in batch culture using a stirred tank reactor with two different initial glucose concentrations (40-50 g/L) and pH values (3.0-4.0) to enhance extracellular polysaccharides production by Pleurotus djamor UNIVILLE 001 and evaluate antitumor effect of intraperitonial administration of Pleurotus djamor extract on sarcoma 180 animal model. According to factorial design, the low pH value (pH 3.0) led to a gain of 1.6 g/L on the extracellular polysaccharide concentration, while glucose concentration in the tested range had no significant effect on the concentration of polysaccharide. With 40 g/L initial glucose concentration and pH 3.0, it was observed that yield factor of extracellular polysaccharide on substrate (Y P/S = 0.072) and maximum extracellular polysaccharide productivity (Q Pmax = 11.26 mg/L.h) were about 188% and 321% respectively higher than those obtained in the experiment performed at pH 4.0. Under these conditions, the highest values of the yield factor of biomass on substrate (Y X/S = 0.24) and maximal biomass productivity (Q Xmax = 32.2 mg/L.h) were also reached. In tumor response study, mean tumor volume on the 21th day was 35.3 cm³ in untreated group and 1.6 cm³ in treated group (p = 0.05) with a tumor inhibition rate of 94%. These impressive results suggests an inhibitory effect of P.djamor extract on cancer cells.

ABSTRACT

Pleurotus sajorcaju MTCC-141 procured from Microbial Type Culture Collection Centre and Gene Bank, Institute of Microbial Technology, Chandigarh has been used for color removal from paper mill effluent. The paper mill effluent amended with basal medium supports the growth of Pleurrotus sajorcaju and removes the colour. The optimum concentrations of carbon source (glucose) and nitrogen source (NH4NO3) for the maximum decolourization of paper mill effluent were found to be 1% and 0.2% respectively. During the fungal growth process, the pH of the paper mill effluent decreased from 7.94 to 4.0.

ABSTRACT

Feeding value of water hyacinth biomass colonized by three species of white rot fungi during solid-state fermentation was investigated. All three organisms proved to be efficient degraders and enhanced dry matter digestibility. Loss of organic matter was maximum (23.6+/-0.1% dry wt) after 48 days by P. ostreatus. C. indica showed maximum cellulose degradation (18.5+/-0.1% dry wt) than other two fungi after 48 days of incubation. In all cases, an extensive removal of hemicellulose at the initial growth period and a delayed degradation of lignin were observed. Hemicellulolysis was maximum (46.3+/-0.1% dry wt) by C. indica, but delignification (14.2+/-0.2% dry wt) by P. sajor-caju after 48 days. The amount of reducing sugar in the degraded biomass decreased at early stages, but increased as degradation progressed in all three cases (maximum 1.1+/-0.05% dry wt after 48 days by C. indica). Soluble nitrogen content increased only during 16-32 days of incubation (highest 1.1+/-0.1% dry wt after 32 days by P. sajor-caju). Crude protein of the bioconverted biomass increased gradually up to 32 days but decreased thereafter (maximum 10.3+/-0.1% dry wt after 32 days by P. sajor - caju). Per cent change in in vitro dry matter digestibility of degraded substrates enhanced gradually after 8 days and reached maximum after 32 days but thereafter decreased (highest + 20.4+/-0.3% dry wt by P. sajor-caju). The results demonstrated the efficient degrading capacity of the test fungi and their potential use in conversion of water hyacinth biomass into mycoprotein-rich ruminant feed, more so by P. sajor-caju.

ABSTRACT

Carbon, hydrogen and nitrogen analyses of banana leaf and pseudostem biomass revealed their potentiality as substrates for microorganisms. Infra-red (IR) spectra of both biomass show presence of cellulose, xylan and lignin. IR spectra of leaf and pseudostem biomass degraded in solid state fermentation (SSF) by two Pleurotus species (P. sajor-caju and P. ostreatus) for 40 days showed the utilization of cellulose, xylan and lignin by these microbes. Dynamics of various lignocellulolytic enzymes of Pleurotus species and analyses of carbon, hydrogen and nitrogen contents of degraded biomass supported the same. Both the Pleurotus species exhibited lignin consumption ability on both the substrates.

ABSTRACT

Pseudostems and leaves from banana waste were used for biotransformation into protein by using P. sajor-caju, an oyster mushroom. Treatment of formalin (500 ppm) + carbendazim (12.5 ppm) of these substrates was found to favour relatively high percentage biological efficiency (BE) of P. sajor-caju.. Steam sterilization also exhibited comparable yield performance by P. sajor-caju. Fruiting bodies harvested from all the treatments had relatively higher protein contents. The spent substrate (steam sterilized) was found to be suitable as an ideal animal feed because of its rich nutritive composition.

ABSTRACT

A modified method for direct determination of cellulolytic activity using Avicel colored with Remazol Brilliant Blue R (RBBR) in Agar test tubes in discussed. Refinaments were introduced in a simple method for qunatitation of cellulase activity, based on the release of dye from Avicel-RBBR medium by enzimatic hydrolysis. Modification in Avicel-dye preparation were enhanced and a spectrophotometer improved the precision of the collected data, since absorbance measurements could be done at the maxmum wavelenght for RBBR (595 nm).
